Severe asthma is a frustrating lung disease marked by destructive lung inflammation that is resistant to steroids, a mainstay of eosinophilic asthma therapy. The inflammation in severe asthma is driven by neutrophils, a type of white blood cell. Researchers have developed effective treatments against eosinophilic asthma by using animal models. Studying severe asthma in the lab has been much more difficult, resulting in few therapies and poor outcomes for patients worldwide. We recently developed a novel mouse model that closely mimics human severe asthma, and we identified an unconventional T cell subset residing within asthmatic airways driving severe asthma. While a pathogenic role for these T cells in allergic lungs is clear, their origin, unique features or link to steroid resistance remains unknown. Our proposed studies will tackle these knowledge gaps of clinical and translational relevance to inform strategies for prevention, control, or recovery from severe asthma.
